A: ELISA quantification of different m6A amounts. Mouse anti-m6A (clone 212B11) was used as capture antibody and biotinylated oligonucleotides served as antigens. Oligos were composed of 24 nucleotides including five N6-methyl-2-deoxyadenosines (m6A oligo BT) or deoxyadenosines (negative control oligo BT) and were biotinylated at their 3′-ends. Streptavidin-conjugated HRP was used as detector. 

B: Release of antibody-bound m6A. Mouse anti-m6A (clone 212B11) was used as capture antibody and was saturated with biotinylated m6A oligonucleotides. Different amounts of free m6A (Sigma) were added to compete with bound oligos. Streptavidin-conjugated HRP was used as detector.
